Description
PTD-DBM is a synthetic, cell-penetrating peptide engineered to interfere with the interaction between CXXC5 and Dishevelled (Dvl), a negative regulator of the Wnt/β-catenin pathway. By blocking this protein–protein interaction, PTD-DBM re-activates Wnt signaling in skin and follicular cells — driving processes associated with hair-follicle activation, wound-induced follicle neogenesis, dermal regeneration, and extracellular matrix remodeling. In preclinical studies, topical PTD-DBM promoted hair regrowth in mouse models and enhanced indicators of follicular proliferation and collagen production in skin cells.
Key Research Applications (hair, pigmentation & skin)
-
Hair follicle activation & neogenesis: Restores Wnt/β-catenin signaling in dermal papilla and epidermal stem cells, promoting re-entry of dormant follicles into anagen and enabling wound-induced follicle formation in rodent models.
-
Support for pigmentation research:Â Because Wnt signaling modulates melanocyte behavior, PTD-DBM is useful in mechanistic studies investigating melanogenesis, follicular melanocyte activation, and pigmentary responses during follicle regeneration.
-
Skin structure & repair: Enhances markers of dermal remodeling (collagen I, α-SMA) and proliferation in fibroblast/keratinocyte assays, supporting studies on skin repair, elasticity, and anti-aging pathways.
-
Combination strategies: Demonstrated synergy with Wnt pathway activators (e.g., small molecules that inhibit GSK3β) to further augment follicular regeneration and wound healing in preclinical work.
Sequence & Chemical Data
-
Core design:Â Protein transduction domain (PTD) fused to a Dishevelled-binding motif (DBM).
-
Representative sequence (research formulations may vary):Â YGRKKRRQRRR-EQLLTRPPTQPKKÂ (PTD linked to DBM sequence; consult COA for exact lot sequence)
-
Molecular Weight:Â ~2.3 kDa (varies by exact construct/modifications)
Recommended Usage (research)
-
In Vitro (cell culture): 0.1 – 10 µM (titrate to assay and cell type).
-
Topical / Ex-Vivo Skin Models: exploratory formulation ranges 0.001% – 0.05% w/v as a starting point for ex-vivo or topical penetration studies — optimize vehicle, pH, and permeability enhancers per formulation.
-
In Vivo (Rodent Models):Â topical application or local injection per published protocols (studies typically use topical dosing schedules; follow institutional animal-care guidelines).
-
Solubility:Â Soluble in sterile water or PBS; vortex gently and avoid prolonged heating. Prepare working dilutions immediately prior to use.
-
Notes: Because PTD-DBM acts via modulation of Wnt signaling, include appropriate controls (Wnt reporters, β-catenin readouts) and validate downstream effects in each model. Avoid extrapolating preclinical results to clinical claims.
Packaging & Storage
-
Packaging:Â 10 mg amber glass vial with desiccant.
-
Shelf Life:Â Up to 12 months unopened (validate with COA).
